Daily cleaning and maintenance of Micropipette
In order to ensure the accuracy and accuracy of the pipette, regular maintenance should be carried out according to the specific use. Especially after using corrosive solvents, the pipette should be cleaned. Through these simple cleaning and maintenance methods, the service life of the pipette can be appropriately prolonged.
L. External cleaning
Method: According to the usage, it can be washed with detergent or disinfected with 60% isopropanol, then washed with double steaming water and dried.
2. Internal cleaning
Generally, the seal ring does not need maintenance. After the piston is cleaned or replaced, a thin layer of silicone grease is coated on the pipette. After the pipette is used up every day, it should be rotated to the maximum scale so that the spring can be restored to its original shape, which helps to maintain the spring elasticity. For liquids of different properties, there are different cleaning and maintenance methods, as shown in the table.

Water solution and buffer are calibrated with distilled water. The result is very accurate. Open the pipette and wash the polluted part with double steaming water. It can be dried in the drying box at temperatures not exceeding 60~C. Apply a small amount of lubricating oil to the piston
Inorganic acid/alkali. If high concentration of acid/alkali solution is often removed, the lower half of the pipette can be occasionally cleaned with double steaming water. It is best to use a suction nozzle with a filter element. Most of the plastic materials and ceramic pistons used in the pipette are acid and alkali resistant materials (except hydrofluoric acid). However, the vapor of acid/alkali solution may enter the lower part of the pipette, affecting its performance. The cleaning method is the same as the "water solution" part.
Potentially contagious liquids. To avoid contamination, a suction nozzle with a filter element should be used. Or use forward displacement method to remove the contaminated part for 121-C, 20-minute high-pressure sterilization, or immerse the lower part of the pipette into the routine disinfectant in the laboratory. Then it is cleaned with double steaming water and dried with the above method.

Cell culture. In order to ensure sterility, a suction with a filter element should be used, referring to the "potentially contagious liquid" cleaning method.

Organic solvents are not allowed to adjust the pipette because of the different density of water; because of the change of vapor pressure and wetting behavior, the pipette should be moved quickly; after the pipette is finished, the pipette should be removed to allow the liquid to volatilize. Normally, for the liquid with high vapor pressure, the natural evaporation process is enough, or the lower part of the disinfectant should be immersed (to ensure that the liquid surface of the immersion does not exceed the position of the sealing ring spring, so as to avoid it. In order to avoid contamination, the radioactive solution should be cleaned by double evaporating water and dried by the above-mentioned drying method.
In order to avoid contamination, nucleic acid / protein solutions should be provided with filter cartridges, or using positive displacement methods: protein disassembly, cleanup and drying methods, such as nucleic acid: Boil 10min in aminacetic acid / hydrochloric acid buffer (pH2.0), ensure that agarose gel electrophoresis does not detect DNA residue, and clean it with double distilled water. The above-mentioned drying method is dry and a small amount of lubricating oil is applied to the piston at the same time.
3 disinfection
It includes ultraviolet disinfection and high pressure disinfection. If the pipette is made of ultraviolet resistant high quality material, ultraviolet disinfection can be used. High-pressure disinfection can be divided into high-pressure sterilization and half-branch high-pressure sterilization of whole pipette. Before the whole high-pressure sterilization and disinfection, the connecting parts should be loosened and partially disassembled and half of them sterilized under high-pressure. The lower part can be disassembled and sterilized under high-pressure (121 C, 20 min). If necessary, the sterilized parts should be dried at room temperature, and then installed after the temperature is completely lowered.
